Aims of the course:Based on our knowledge of genetics and molecular genetics to acquaint students with traditional and modern breeding methods and procedures point to the potential use of genetic varieties praxi.Second part of the course focuses on using the latest molecular techniques and procedures in practical plant breeding .
Course contents:
1.Introduction and basic concepts of classical plant breeding. (allowance 4/0)
 
a.The status of plant breeding in agriculture, the importance of breeding, history and development of plant breeding in the world and us. Genetic structure of the variety, variability of cultivated plants, important cultural properties and characteristics of flora
b.The main phases of the breeding process, genetic resources, centers of origin of cultivated plants, preservation and conservation of genetic resources. Implications for the propagation of breeding methods and selection procedures, techniques and procedures for crossing

2.Conventional techniques, methods and practices of breeding. (allowance 6/0)
 
a.The techniques and selection methods. Breeding methods for self, for cross-pollinated, in vegetatively propagated. Peculiarities of the biennial and perennial species.
b.Nature of heterosis, heterosis theory explaining, using heterózního effect in plant-breeding techniques for the breeding of F1 hybrids. Male sterility, genetic determination of male sterility, the use of male sterility in breeding F1 hybrids

3.Alternative breeding techniques (allowance 6/0)
 
a.Mutation breeding, induced mutagenesis, mutagens used, methods of working. Remote hybridization causes problems with pollination of species and the possibility of overcoming, the properties of distant hybrids
b.Properties of polyploids, the use of polyploidy in plant breeding, methods of obtaining polyploid breeding, use of aneuploidy. haploid and in plant breeding. Breeding for resistance to pests and diseases, genetic nature of resistance

4.Molecular markers, their nature and use (allowance 6/0)
 
a.Hybridization techniques used to detect molecular markers. RFLP, VNTR, FISH and more
b.Techniques based on polymerase chain reaction (PCR): RAPD, SSR, AFLP
c.Methods for DNA sequencing
d.The use of molecular markers to analyze genetic resources (genotyping) and using selection markers (marker assisted selection, MAS)

5.Gene manipulation in plant breeding (allowance 6/0)
 
a.The basic strategy of gene manipulation in plants, gene cloning and cloning vectors, expression vectors
b.The use of Agrobacterium tumefaciens in Transgenosis plants, other technologies can obtain genetically modified plants.
c.Objectives Transgenosis in plants, most frequently used genes, characteristics of the GMOs, placing GMOs on the market.
d.European and Czech legislation governing the handling of GMOs

-Ability to compile a methodology for the experiment, implement the experiment and evaluate it
-The ability to draw conclusions applicable in breeding from the results of molecular analysis
-The ability to propose an approach for the desired properties of the plants with the classical and biotechnological methods.
-The ability to propose how to reach the projected plant traits by means of classical and biotechnological methods.
-The ability to select a suitable molecular marker for the plant species for the MAS (Marker assisted Sellection)
